1. Wipe the injection port with 70% isopropyl alcohol, let evaporate for 10 seconds
2. Inject 2β5 ml of liquid culture, spore suspension, or agar transfer (18β21G needle)
3. Incubate in the dark at 23β25Β°C (73β77Β°F) in a clean environment
4. Perform shake and break when colonization reaches 30β50% (usually 7β10 days)
5. At full colonization (10β14 days total), inoculate bulk substrate (straw, supplemented sawdust, etc.)
